To assess the effects of this legislation, the paper will consider several studies that focus on the financial impacts brought about by disclosure of financial information then engage in an independent discussion on the same (DeFond, 2004). Analysing isolation of DNA plasmid and Agragose of gel electophoresis Introduction (a) The aim of this experiment was to successfully isolate a DNA plasmid from E.Coli cells (Escherichia coli). We then use commonly performed a method commonly used in biochemistry and molecular biology called agarose gel electrophoresis. This is used to separate DNA and RNA fragments according to length are used to estimate the size and charge of the DNA and RNA fragments or to separate protein by size. In this procedure as stated above, we used e.coli as these are plasmid containing cells. These cells were placed in a buffer and mixed with a solution of 1% (w/v) SDS (sodium dodecyl sulphate) which was mixed with sodium hydroxide. The alkaline solution (12.6PH) causes the molecular weight increases this causes it to become like chromosomal DNA. Using alkaline lyses is based on differential denaturation of chromosomal and plasmid DNA in order to separate the two. The double stranded plasmid and chromosomal DNA is converted to single stranded DNA due to the lyses of the cells which solubilises protein and denatures the DNA. Subsequent neutralization is potassium acetate allows only covalently closed DNA plasmid DNA to reanneal and stay solubilized. Chromosomal and plasmid DNA precipitate in a complex formed with potassium and SDS which is removed by centrifugation. Protein dodecyl sulphate complexes are precipitated die to it being insoluble in water. When centrifugation neutralizes the lysine it yields to a minuscule supernatant fraction that contains plasmid DNA a network of chromosomal DNA and protein Plasmid DNA is concentrated by from the supernatant by ethanol precipitation. Plasmid DNA isolated by alkaline lyses is suitable for most analyses and cloning procedures without further purification however if the isolated plasmid DNA is sequenced and additional purification step such as phenol extraction is used. (b) The aim of Agarose gel electrophoresis is to analyse the plasmid DNA that was extracted from the procedure before. The technique of electrophoresis is based on the fact that DNA is negatively charged at neutral pH due to its phosphate backbone. And like any other biological macromolecules can move within an electrical field. The rate of the DNA slows down when its moves towards opposite poles because of the agarose. The agarose gel is a buffer solution this is used to maintain the required pH and salt concentration. The agarose forms hole or wells in the buffer solution and the DNA inserted in through the holes to move toward the positive pole. As mentioned before the agarose gel slows down the rate of DNA so the smaller DNA moves faster than the larger molecules of DNA as the smaller ones fit through the whole easier. This causes the DNA to be separated by size and can be seen visually. To make the electrophoresis to function and separate DNA molecules it must contain an electro phoresis chamber.and power supply, combs which are placed in the chamber this is how wells are formed when agarose is placed in the gel, Trays that contains a special gel that comes in many sizes and and have UV-properties combs which is how wells are formed when agarose is placed in the gel, Electrophoresis buffer, Loading buffer, which has a thick consistancy (e.g. glycerol) so the DNA can be easily placed in the wells and one or two tracking dyes, these travel in the gel and help visualize how the process is being carried out and to moniter how far electrophoresis undergone. Ethidium bromide, is a dye used to stain the nucleic acids.  . Tran illuminator  (an ultraviolet light box), which is used to visualize ethidium bromide-stained DNA in gels.   Method for plasmid isolation 1.5 ml of culture that contains E.coli cells containing the plasmid pUC118 was inserted into an Eppendorf tube. This was then centrifuged at 13000 rpm for two minutes The liquid contained in the Eppendorf tube was discarded carefully by using a pipette and then inverting the tube on a test tube to remove remaining drops of the liquid without removing the bacterial pellet 200 micro-liters of solution A was added to the bacterial pellet. This ensured that the suspension is homogenized (mixtures are well separated 400 micro-liters of solution B was then added and mixed well these solutions contain the SDS and sodium hydroxide. This neutralizes the solutions 300 micro-liters of solution C which contains the potassium acetate which was also mixed and then was incubated on ice for 10 minutes This mixture was the centrifuged at 13000rpm for 5 minutes 750 micro-liters of this supernatant was transferred to a new Eppendorf tube whilst ensuring none of the precipitate was interfered with 10 micro-liters if RNAse solution was added to a duplicate tube and labeled as R+ 450 micro-liters of isopropanol was added to each test tube and mixed well This was then centrifuged at 13000rpm for 5 minutes The supernatants were then carefully removed and the DNA was retained 400 micro-liters of ethanol was added and allowed to stand for a minute it allow the salts to dissolve the liquid was carefully removed so as not to remove the DNA precipitate. The sample was then allowed to dry at room temperature Each pellet was then dissolved in 10 micro-liters of TE buffer Q1 The viscosity after 400 micro-liters of solution B was added and mixed a low viscosity was observed as it had a very watery texture. Q2 there was no viscosity after the transfer of 750 micro-liters of supernatant to a new eppendorf (a) Agarose gel electrophoresis The sample obtained from the experimental procedure above were then examined using the method of agarose gel electrophoresis The RNAse treated and untreated plasmids were examined. 10 micro-liters of loading buffer was added to 10 micro-liters of DNA for each sample The samples containing DNA mixed with loading buffer were then pipetted into the sample wells, and a current was applied. This was carried out for 30 minutes It was clear that the current was flowing as bubbles were observed to be coming off the electrodes. The negatively charged DNA migrated towards the positive electrode at the distal end, (which is usually colored red) It was analyzed that the smaller DNA molecules travelled quickly through the gel which showed that the procedure was carried out successfully as the DNA was separated according to size Results/ Discussion (a) Isolation of DNA plasmid The DNA plasmid was successfully extracted from the E.coli cells and then the DNA was the successfully separated according to size by using the agarose gel electrophoresis method. Solution A contains 25 mM of Tris-HCL (pH 8.0)50 EDTA. Tris is a buffering agent this maintains a constant pH. The EDTA is used to protect the DNA from DNAses which are degradative enzymes; the EDTA also binds divalent cations that are necessary for DNAse activity. The solution B contains SDS which is a detergent and NaOH. This neutralizes the solution, the alkaline mixture also causes the cells to rupture and the SDS the lipid membrane is broken apart and the cellular proteins are solubilized, NaOH converts the DNA into a single strands which is caused by denaturation. The solution C contains potassium acetate (pH 4.3) the acetic acid neutralizes the pH, allowing the DNA strands to renature. The potassium acetate is added its causes the SDS to precipitate, along with the cellular debris. The  E. coli chromosomal DNA is also precipitated. The plasmid DNA remains in the solution. The viscosity of this is very high as it has a very gel like texture. When the supernatant is placed in a new eppendorf tube after 5 minutes of centrifuge this causes the plasmid DNA to separate from the cellular debris and chromosomal DNA in the pellet. The isopropanol is then added this pulls the plasmid out and causes it to precipitate nucleic acids. After centrifuge a small white pellet was observed at the bottom of the tube after the supernatant was carefully removed this further purifies the plasmid DNA from contaminants. 400microliters of ethanol was added this washed the residual salt and SDS from the DNA. All these changes that were observed after the addition of these solutions were expected as they are what help us extract the DNA plasmid for an end product. (b) Agarose gel electrophoresis After placing the DNA plasmid in the wells electrophoresis was carried out. The results were then obtained and recorded. The size of the DNA fragment is determined from its electrophoretic mobility. The DNA fragments of know molecular weight markers are run on the gel and a graph of log MW against migration distance is drawn. There are three different forms of agarose DNA first theres the open circular plasmid DNA this is the first band that occurs on the picture. The circular plasmid is a  double-stranded  circular  DNA  molecule  that has been nicked in one of the strands to allow the release of any super-helical turns present in the  molecule. The open circular plasmid migrates more slowly than a linear or super-coiled  molecule  of the same size this is due to associated differences in  conformation, or shape, of the  molecules. this is why it is the first band that occurs on the picture result. Linear DNA has free ends, either because both strands have been cut, or because the DNA was linear  in vivo. The rate of migration for small linear fragments is directly proportional to the voltage applied at low voltages. At a specified, low voltage, the migration rate of small linear DNA fragments is a function of their length. Large linear fragments (over 20kb or so) migrate at a certain fixed rate regardless of length. This is because the molecules resperate, with the bulk of the molecule following the leading end through the gel matrix.  Restriction digests  are frequently used to analyse purified plasmids. These enzymes specifically break the DNA at certain short sequences. The resulting linear fragments form bands after  gel electrophoresis. It is possible to purify certain fragments by cutting the bands out of the gel and dissolving the gel to release the DNA fragments. This is neither fast nor slow in comparison to the other DNA plasmid. The super-coiled Plasmid DNA normally occurs naturally, there is super-coiling in DNA only if there is a replication of a DNA plasmid and this occurs for a small space of time and that is removed by cutting the DNA by specific enzymes, this is part of DNA replication mechinary. This type of DNA plasmid is the fastest as it is the last band shown out of the three this is Because of its tight conformation. The picture above shows the results obtained from the agarose gel electrophoresis. The lane numbers are marked over the wells. The lane before lane 1 that is titled M is the molecular weight marker. All three forms of plasmid DNA is present in this result, the open circular, the linear and the supercoiled. There is an extra band of RNA present however not clearly visible this is because the RNA fragments migrated ahead of dye front as diffuse a band, the ribonuclease gets rid of this band, a blue tracking dye cause the black smudge under the DNA plasmid and beneath that is the barley visable RNA. RNA is very unstable under these conditions, as a result RNA can be completely degraded befor the ribonuclease has been added. It can be seen that DNA is present more in one band then another, however the one with the less amount could have a bigger fragment. There seems to be logarithmic relationship between the size of the DNA fragment and the distance it travels on the gel. A standered curve can be made if we measure the length the bands in different lanes travelled if the fragment sizes are known. The more points plotted and the larger the separation there is on the gel, the results will be more accurate. Conclusion The experimental procedures carried out were a success, the DNA plasmid was obtained and the agarose gel electrophoresis resulted with in a clear picture as shown and outlined above, of the DNA being successfully separated. The uses of purified plasma in DNA research is for molecular cloning.

Energy is neither created nor destroyed. Energy is summed up into two different properties: Potential energy and Kinetic energy. If potential energy increases then kinetic energy decreases and vice versa.   Ã‚  Ã‚  Ã‚  Ã‚  In this experiment we positioned a marble ball on a wooden roller coaster positioned on a physics stand in the sixth hole. Throughout the experiment, we used an electronic timer to record the time of the marble where it passed through the light beam of its clamp. We positioned the clamp at a certain point on the roller coaster and measured the distance from the marble to the clamp; the height of the clamp; and finally the time the ball traveled through the clamp. After we recorded these different figures we calculated the speed of the marble from the given distance traveled and the time. We repeated the step 14 times, then proceeded to graph the speed and the height. Next, we took the measurements of position of the clamp, height, and speed and calculated the potential energy, the kinetic energy, and the total energy. Total energy calculated as mentioned before. Potential energy is taking the mass (m) which is 28.1g times gravity (g) which is 9.8 m/s2 times the he ight. Kinetic energy is one-half times the mass (m) times velocity (v2). Finally we graphed the calculated kinetic, potential, and total energies of this experiment.   Ã‚  Ã‚  Ã‚  Ã‚  For this theoretical result, the motion or speed of the marble will have different calculations. Also, the potential energy and kinetic energy of the marble will produce a mirror effect between each other respectively. A doll house Essay

Henrik Ibsen play â€Å"A Doll House,† written in 1879, focuses on a story of a disparaging role of women in Victorian society through his doll motif, played out in Nora’s sudden distaste for her home. Throughout the play there are many examples of Nora’s husband Torvald treating Nora in an insulting manner because she’s a woman. Torvald calls her little pet names, and states that she’s frail. Nora does things according to what Torvald wants. Everything is done by his standards. He also doesn’t allow her to have much freedom. He doesn’t let decisions to be made by Nora. Torvald makes comments that suggest Nora could never understand anything, just because she is a woman. These examples show that feminism is a theme throughout the story. Torvald treats Nora almost like a child. He never actually talks to her like an adult. Almost as if Torvald thought that Nora wasn’t intelligent, or mature enough to have a conversation that had to deal with serious matters. He also has a lot of pet names for Nora. Whenever Torvald speaks to   Nora he usually calls her â€Å"my little squirrel†, and â€Å"little lark† as you would call a child. Torvald   also calls her a spendthrift whenever she asks for money. He never really calls her Nora, unless it is when he is serious, however any other time, he will call her by one of his pet names. Torvald also never speaks to Nora about anything important. He only talks to her about spending money, and about things of leisure, like the ball. Nora, in Ibsen’s â€Å"A Doll’s House†, is a modern woman limited by a traditional society which denies women power and autonomy. The central mystery and challenge of â€Å"A Doll’s House† is obviously the character of Nora. The story starts on Christmas Eve. Nora makes preparation for Christmas. While she eats macaroons, Dr. Rank and Mrs. Linde enters. Rank goes to speak with Torvald while Linde speaks with Nora. Linde explains that her husband has died and that she needs to find a job. Nora agrees to ask her husband to give Linde a job at the bank. Nora tells her about borrowing money to pay for the trip they took to Italy. She explains that Torvald doesn’t know that she paid for it. Rank leaves the study and begins to speak with Nora and Linde. He complains about the moral corruption in society. Krogstad arrives and goes to the study to talk to Torvald about keeping his job. A few minutes later, he leaves and Rank comments that Krogstad is one of the most morally corrupt people in the world. Rank and Linde leave, and Krogstad re-enters. He tells Nora to ask her husband to keep Krogstad at the bank, or else. If she doesn’t, he will reveal Nora’s crime of forgery to him. Krogstad leaves and when Torvald re-enters, Nora asks him not to fire Krogstad. Torvald says that he must fire him because of his dishonesty and because he gave Krogstad’s job to Linde. The nurse, Anne-Marie, enters and gives Nora her ball gown. Anne-Marie explains that she had to leave her children to take the job taking care of Nora. Linde returns and begins to help Nora with stitching up her dress. They talk for a while about Dr. Rank. Torvald enters and Linde leaves to the nursery. Nora asks Torvald again not to fire Krogstad and he refuses. He gives Krogstad’s pink slip to the maid to be mailed to Krogstad. Rank re-enters and tells Nora about his worsening illness. They talk and flirt for a while. Rank tells Nora that he loves her. Nora said that she never loved Rank and only had fun with him. Rank leaves to the study and Krogstad enters. He is angry about his dismissal and leaves a letter to Torvald explaining Nora’s entire crime in the letter box. Nora is frightened, and tells Linde about the matter and Linde assures her that she will talk to Krogstad and set things straight. Linde leaves after Krogstad and Rank and Torvald enters form the study. They help Nora practice the tarantella. After practice, Rank and Torvald exits and Linde enters and tells Nora that Krogstad left town, but she left a note for him. Nora tells her that she’s waiting for a miracle to happen. That night, during the dance, Linde talks to Krogstad in Helmer’s apartment. She explains to him that she left him for money, but that she still loves him. They get back together and Krogstad decides to forget about the matter of Nora borrowing money. However, Linde asks Krogstad not to ask for his letter back since she thinks Torvald needs to know of it. Both leave and Torvald and Nora enter from the dance. Torvald checks his letter box and finds some letters and two business cards form Dr. Rank with black crosses on them. Nora explains that they meant that Rank is announcing his death. After the bad news, Torvald enters his study and Nora prepares to leave. However, before she can get out the door, she is stopped by Torvald who read Krogstad’s letter. He is angry and disavows his love for Nora. The maid comes with a letter, Torvald reads the letter that is from Krogstad. It says that he forgives Nora of her crime and will not reveal it. Torvald burns the letter along with the IOU that came with it. He is happy and tells Nora that everything will return to normal. Nora changes and returns, she tells him that they don’t understand each other and she leaves him. Joan Templeton wrote a critical piece of â€Å"The Doll House Backlash: Criticism, Feminism, and Ibsen.† Templeton states â€Å"Whatever propaganda feminists may have made of A Doll House, Ibsen, it is argued, never meant to write a play about the highly topical subject of women’s rights; Nora’s conflict represents something other than, or something more than, woman’s. In an article commemorating the half century of Ibsen’s death, R. M. Adams explains, â€Å"A Doll House represents a woman imbued with the idea of becoming a person, but it proposes nothing categorical about women becoming people; in fact, its real theme has nothing to do with the sexes† (416). Over twenty years later, after feminism had resurfaced as an international movement, Einar Haugen, the doyen of American Scandinavian studies, insisted that â€Å"Ibsen’s Nora is not just a woman arguing for female liberation; she is much more. She embodies the comedy as well as the tragedy of modern life.†Ã¢â‚¬  (28). Joan Templeton had cited â€Å"All female, or no woman at all, Nora loses either way. Frivolous, deceitful, or unwomanly, she qualifies neither as a heroine nor as a spokeswoman for feminism. Her famous exit embodies only â€Å"the latest and shallowest notion of emancipated womanhood, abandoning her family to go out into the world in search of ‘her true identity;† (Freedman 4)† (30). Nora Helmer makes the right decision to free herself form the social and traditional commitments and obligations of the Victorian Era and becomes an independent individual. She lived in a world of pre-determined social and societal constraints that made her deprived of her own freedom and happiness. The society in which she lived wanted people to live according to the rigidly set norms and standards of the Victorian Society. Subjugation and oppression was the theme of the Victorian Society. Men and women were supposed to play the role that was assigned to them. Nora found herself in such a world of suppression. She was supposed to live a quiet life in a world that was dominated by her husband Torvald and the alike. She was however, totally dissatisfied with the life of subjugation. She could no longer surrender to the constraints of the society. The made her brake from the captivity and enters a new world of freedom. Nora Makes the right decision to free herself from the social and traditional commitments and obligations and become an independent individual. Nora is indeed a classical hero during her time of Victorian Society. She was hiding her character and personality throughout the play under the pretense of the ideal 19th century wife who completely abides to her husband. The character of Nora is quite tough to interpret, as she is made out of a combination of different traits, childish, and even selfish. Even though she is found to be playful and silly, she appears different in other places being practical and astute. She is indeed a hero as she was successful in showing that she is a supporting wife, and mother. Nora was expected to be content with the life she had, though it wasn’t in any way fair or equal. When she expresses her hope that Torvald would have taken the blame for her crime upon himself, Torvald says that â€Å"there’s no one who give up honor for love.† (875) and Nora replies that â€Å"millions of women have done just that.†(875). When Nora shut the door behind her, she wasn’t just a woman leaving her family. She was a woman seeking independence from the strictures of society and the rule of men which was placed upon her because of gender. A Doll House Essay Reflective Statement In Ibsen’s A Doll House, the setting is a critical part of understanding the issues presented in the piece. If it had not been set in 19th century Norway, many of the references would not have been possible. The setting of each of the acts the house changes, showing an allusion of a perfect doll house in the first act that is slowly diminishing threw out the last two. As a external way of showing the cultural and emotional conflicts with in the house. Plus, the significance of the cultural statement, about the unfairness of women’s roles, would have been lost. From this, we can conclude that it was relatively easy to understand Ibsen’s view on the social and cultural issues regarding women- a blatant inequality between the genders, both at home and in society. Regarding Mrs.Linde who comes into the play as a woman who has seen the outside world and worked for her living, Ibsen at the end shows her beginning a relationship with Krogstad to sho w even when a woman trails away from the protection of a man they always come back to their original place. It is difficult to understand the solution to these cultural issues, unless all women are supposed to walk out of their lives. If that were the case that would create more and if not equal cultural struggles and issues. This is similar to today, in that we are still trying to work on the inequality in society through efforts like affirmative action, with questionable success. That is why Ibsen’s use of techniques like setting and characterization are so important; these techniques are what Ibsen uses to convey his message to the audience. To show the struggle of each individual relationship in this work whether it be between Nora and Torvald Helmer, Mrs.Linde and Krogstad, or Dr. Rank and his undying love for Nora. A Doll House Essay Marriage, as an institution, facilitates the union of man and woman enabling them to raise a family. The wellbeing and happiness of the family depends equally on the man and the woman. But this equality is imperceptible in married relations, even in the modern society. The woman always compromises for the sake of her family and abandons her freedom. The play â€Å"A Doll House† by Henrik Ibsen depicts the situation of a married woman, Nora Helmer who has to repress her desires and behave according to the wish of her husband, Torvald. But as the play progresses conflicts began to arise between Nora and Torvald, owing to the changing nature of Torvald’s love for Nora. Throughout the play, Nora aims to act and behave in a manner which pleases her husband but at the end of the play, Nora decides to leave Torvald and lead her life in accordance to her wishes. The end of the play depicts a fit resolution to the main conflict in the play. End of the Play Nora is a skilled and wise woman but her husband regards her to be an asinine and childish woman. When Torvald was ill, Nora was the one who saved his life with the aid of money borrowed from Krogstad. But she refrained from revealing about the borrowed money to her husband so that his pride is not hurt. She also worked secretly to repay the debt. But she presents herself before her husband in a manner which pleases him. Her husband thinks that Nora is a silly woman and Nora continues to act like a one before him. Nora lives in her house like a doll whose strings are in the hands of her husband. But in the final scene of the play, Nora goes on to become an independent woman from a doll whose actions were controlled by her husband. Owing to Krogstad’s letters which reveal the details of the bond that Nora has signed while borrowing money from Krogstad and the forging of her father’s signature by Nora on that bond, Torvald suspects Nora’s loyalty towards her family. Torvald even goes to the extent of saying that their marriage has ended. He also prohibits Nora from raising her own children. Ibsen (2002) writes, â€Å"But I shall not allow you to bring up the children; I dare not trust them to you. † (pg 135). But when Torvald receives another letter from Krogstad, consisting of the bond, he is thrilled to be liberated from the bond. Now he praises Nora for her dedication and love that she had for her family, and also mentions that he has forgiven her for her mistakes Nora realizes that Torvald’s love for her is so conditional that it is easily influenced by the situations around them. Torvald is so concerned about his honor that he never thinks about the feelings of Nora. He fails to live up to the role of a husband who is responsible for protecting his wife’s honor. Nora expresses her discontent at Torvald’s self-centered behavior and decides to leave the house. Instead of being just a wife and mother, Nora desires to become a sensible human being. Ibsen (2002) writes, â€Å"I believe that before all else I am a reasonable human being. Just as you are—or, at all events, that I must try and become one. † (pg. 145). Torvald tries to convince Nora to stay with him but Nora refuses to listen to him. She leaves the house and goes in pursuit of an independent life where she can live according to her own desires and wishes. The end of the play serves as an appropriate resolution to Nora’s conflict with Torvald. Nora refuses to lead a doll-like life in order to save her marriage from breaking up. Language and Culture Essay

Language and culture reciprocate a fluid relationship. They both interact and shape the structure through which individuals can mediate their lives within a social context. Language and culture are immanent forces that help to define and open up the way we understand various dimensions of our lives, whether through the mass media and advertising, science and technology, slang, diverse vocabulary, changes of meanings. This essay will aim to break down the specific forums through which language is useful to culture and how culture forms the mechanisms and strategies through which we use language. I will begin with a discussion of the relationship between language and culture and then move on to the different paradigms that contain specific uses of language. To begin, it will be helpful to lay a framework upon which to build a working definition of language. According to the University of Princeton’s online dictionary, language is â€Å"a systematic means of communicating by the use of sounds or conventional symbols† (Language). This is straightforward enough for us to understand that language acts as a medium for communicating. The key terms to highlight here are systematic as well symbols because they stretch the function of the definition to include the power that institutions have over language as well as the way that language can shift and change through semiotics. As a system, language relies upon a specific and formal set of rules in order to function. Grammar, syntax, slang, and meaning are all elicited from the rules of language. The institution of a particular language also varies greatly depending on socio-economic factors as well as through educational strategies. For example, with the rise of information technology and the internet as well as through global capitalism, the English language has taken center stage as a pseudo-global language due to its far reaching capacity and its prevalence in scientific and technological innovations. It will be interesting to see how the role of English changes as the world becomes smaller through interconnectivity and the rise of other global languages such as Chinese. Different cultures, through different languages, create different modes of expression and understanding that give rise to cultural diversity. Cultural diversity relies upon the comparing and contrasting mechanisms through which meaning is ingrained. Semiotics is the realm of language that is concerned with signs and signification. Language is the tool through which we identify particular signs and accord them a meaning, or a signification. This is important because through the sign, or symbol, a culture places specific connotations beyond the traditional denotative structure of language. Language both identifies and distinguishes. This is where varying mediums play a significant role on the way cultures digest and create their lives through language. To illustrate how this works, I will next discuss the role that the mass media has on both the language of culture and the culture of language. The dominance of mass media and advertising over the realms of cross cultural communication cannot be underestimated. Through advertisements, newspapers, television, radio, and the internet, people across the world have instant access to a constant stream of information that shapes the way we live our lives. This can have both positive and negative impacts. On the positive side, mass media acts a vehicle for cross cultural dialogue. It provides people with a common language and a common set of terms through which to discuss the pressing issues of the day. The information presented is current and has a specific point of focus. Depending on a person’s cultural condition and through which avenue of mass media they travel on the information highway, the meaning of the information changes dramatically. In this way, two people from different cultures can watch the same news clip and come away from the experience with two completely different understandings from the same language and information. On the negative side of mass media we can point to what cultural theorists beginning with Antonio Gramsci have termed cultural hegemony. This names a dual process through which competing ideologies are struggled out on the stage of culture and through which subversive or outsider paradigms are brought into the dominant arena of culture in order to assimilate and therefore neutralize the conflict of interests (Chandler). This applies to our discussion of language and culture because it helps explain how the role of the media frames certain issues in order to parlay a particular point of view. For example, here in America hegemony can be witnessed through the way Fox News presents its conservative political agenda against the way that Comedy Central portrays its progressive politics through shows like the Daily Show. The language presented and used in media relies upon subtle ideological functioning in order to suggest at an agenda or viewpoint instead of simply communicating said purpose. As mentioned before, language both identifies and distinguishes. As in the previous example of news presentation, we can see how Fox News identifies its audience through its conservative ideology. By identifying as they do, they also distinguish themselves from the liberal-leaning Daily Show audience. It will be interesting to see how much, if any, influence Fox News can have during the Obama presidency and the rise of liberalism after relying so heavily upon Republican and conservative politics through the Bush administration. The shift in the power of balance in American politics will sway the tide of public opinion in the arena of culture that hegemony frames. We can also witness the way science and technologies rely upon specific uses of language in order to elucidate communication and meaning. Perhaps this arena of culture illustrates the example best. Science and technology create paradigms of knowledge. By this, we can see how biochemists almost literally speak another language than nuclear physicists in their professional lives. Again, this is not a positive or a negative situation; it is a cultural practice that plays itself out through particular frameworks of understanding the world. Another interesting example of how science and technology play out in the realm of language is to consider the macro level. As hinted at earlier, science and technology, with their innovations originating largely from the United States and Japan, have consisted and evolved through the language of not just professional jargon, but specifically and nearly exclusively through English. In his innovative essay, Translingual Travel: The Discourse Practice of Cultural Hegemony, Chinese cultural theorist Dai Xun writes of the impact this phenomenon has in China, â€Å"The primary premise in the rise of cultural hegemony is the advantages and control western countries enjoy over science, technology and information (Xun). This is another form of cultural hegemony that phases out periphery languages at the cost of integrating English into the global vocabulary as well as forming the unbalanced socio-economic relationships of our age. Language is utilized for specific purposes through systematic controls and symbolic gestures. That being said, language is not a monolithic and static entity. Although language relies on specific grammatical and syntactical rules, cultures have always innovated and bent the rules of grammar in an effort to assert their unique cultural conditions. The rise of slang is one of the most important and culturally reinforcing tactics that marginal groups use to coalesce and self-identify. Slang is a part of language, and it follows that it serves again to identify as well as to distinguish. One of the most striking examples of slang can be witnessed in hip-hop music. Hip-hop is rapidly becoming a global phenomena and it goes beyond just the music to include fashion, dancing, music making and lifestyle. In this way, the slang the hip-hop community uses separates them from other groups while also helping them to identify with like-minded people. Hegemony in the realm of hip-hop can help explain how power relations work themselves out through culture. In its infancy, hip-hop was a culture unique to African-Americans in inner-city America. With its popularity and rise through mass media, white, middle-class teenagers are adopting hip-hop into their lifestyles and trying to identify with the conditions that gave rise to it. This is how dominant culture integrates what was previously a subversive and politically threatening subculture into the mainstream. Now we see hip-hop artists in children’s cartoons, at suburban schools and in the malls. In conclusion, language and culture maintain mutually reinforcing relationship. Both dominant and subversive forms of communication are played out on the arena of cultural hegemony. This condition goes beyond the traditional positive/negative paradigm of culture. First, they hypothesized that certain personality traits would be perceptually related to a certain hair color: that the participants would find blondes more attractive, feminine and immature; brunettes more intelligent, successful, mature and stronger work ethics; and redheads more aggressive and emotional. Second, they hypothesized that the male and female models with brown hair would be viewed more favorably in the work setting than in the date setting. Third, it was hypothesized that the typical stereotypes associated with hair color would become stronger when combined with situations common to these stereotypes. classism and the criminal essays While searching the web for articles on classism, I had the fortune to come across a fascinating site called the Anarchist Black Cross Network. What captured my attention was a speech by Clarence Darrow to the inmates in the Chicago prison in 1902. Being impressed by his philosophy, I determined to learn as much as I could about who Darrow was and what he did. During the process, I have come to appreciate Douglas O. Linder for his article, Who is Clarence Darrow, (1997). Clarence Darrow was a political activist and trial lawyer. Linders article summarizes Darrows famous speech to the inmates, which highlights his philosophy and also covers some of his more famous trials. Darrows main belief is that there is no such thing as a criminal; society causes people to exist in whatever manner they do, that is whether they are inside of or outside of a prison. He states to the inmates that most people outside of prison are criminals in an acceptable way i.e. the gas company, retailers, etc. When Darrow looks at advertisements all he sees are lies. Darrow views selling for profit as a societally acceptable crime. I believe that Darrow views himself as a Robin Hood of sorts, exposing the richs exploitation of the poor as crime. He was a visionary for his time: reading his text, I felt as if the views were very modern and could have been written recently. Darrow expresses that humanity is forced into crime because of poverty. He gives examples. The first is kidnapping. Darrow states, This crime is born, not because people are bad; people dont kidnap other peoples children because they want the children or because they are devilish, but because they see a chance to get some money out of it. Court Transcript of BTKs Confession On February 26, 2005, the  Wichita Police announced that investigators had made an arrest in the BTK serial killer case after taking into custody an employee of nearby Park City, Kansas in a routine traffic stop – bringing to an end an era of terror for the Wichita community which lasted more than 30 years. Dennis Rader,  a city employee, a cub scout leader, and an active member of his church, confessed that he was the BTK serial killer. Here is the transcript of his confession. The Defendant: On January 15th, 1974, I maliciously, intentionally and premeditation killed Joseph Otero. Count Two – The Court: All right. Mr. Rader, I need to find out more information. On that particular day, the 15th day of January, 1974, can you tell me where you went to kill Mr. Joseph Otero? The Defendant: Mmm, I think it’s 1834 Edgemoor. The Court: All right. Can you tell me approximately what time of day you went there? The Defendant: Somewhere between 7:00 and 7:30. The Court: This particular location, did you know these people? The Defendant: No. That’s –(Off-the-record discussion between the defendant and Ms. McKinnon.) No, that was part of my – I guess my what you call fantasy. These people were selected. The Court: All right. So you – (Off-the-record discussion between the defendant and Ms. McKinnon.) The Court: – you were engaged in some kind of fantasy during this period of time? The Defendant: Yes, sir. The Court: All right. Now, where you use the term â€Å"fantasy,† is this something you were doing for your personal pleasure? The Defendant: Sexual fantasy, sir. The Court: I see. So you went to this residence, and what occurred then? The Defendant: Well, I had – did some thinking on what I was going to do to either Mrs.  Otero or Josephine, and basically broke into the house –or didn’t break into the house, but when they came out of the house I came in and confronted the family, and then we went from there. The Court: All right. Had you planned this beforehand? The Defendant: To some degree, yes. After I got in the house it – lost control of it, but it – it was – you know, in back of my mind I had some ideas what I was going to do. The Court: Did you – The Defendant: But I just – I basically panicked that first day, so – The Court: Beforehand did you know who was there in the house? The Defendant: I thought Mrs. Otero and the two kids – the two younger kids were in the house. I didn’t realize Mr. Otero was gonna be there. The Court: All right. How did you get into the house, Mr. Rader? The Defendant: I came through the back door, cut the phone lines, waited at the back door, had reservations about even going or just walking away, but pretty soon the door opened, and I was in. The Court: All right. So the door opened. Was it opened for you, or did someone – The Defendant: I think one of the kids – I think the Ju – Junior – or not Junior – yes, the – the young girl – Joseph opened the door. He probably let the dog out ‘cause the dog was in the house at the time. The Court: All right. When you went into the house what happened then? The Defendant: Well, I confronted the family, pulled the pistol, confronted Mr. Otero and asked him to – you know, that I was there to – basically I was wanted, wanted to get the car. I was hungry, food, I was wanted, and asked him to lie down in the living room. And at that time I realized that wouldn’t be a really good idea, so I finally – The dog was the real problem, so I – I asked Mr. Otero if he could get the dog out. So he had one of the kids put it out, and then I took them back to the bedroom. The Court: You took who back to the bedroom? The Defendant: The family, the bedroom – the four members. The Court: All right. What happened then? The Defendant: At that time I tied ‘em up. The Court: While still holding them at gunpoint? The Defendant: Well, in between tying, I guess, you know. Analysis On The Movie Inception Film Studies Essay The film Inception (2010) captured the imagination of many spectators as one of the best science fiction and fantasy movies to be produced that year. It was produced and directed by Christopher Nolan who also wrote the script of the movie. The development of this script and the story line were Nolans original ideas back in 2001, nine year before the movie was released when he wrote an eighty-page script on dream stealing. This work was inspired by the concept of dream incubation and lucid dreaming which bore this mystery envisioning a world where technology has been developed to allow entrance into the human mind by means of dream invasion. Inception is based on the basic inspiration that a single idea in an individuals mind can be either the most valuable asset or the most dangerous weapon (George, 2009). The film stars Leonardo DiCaprio as Dom Cobb, who in the movie steals information from the minds of his victims during dreams. The last job which Cobs is given, and which is the ce nter of the movie involves performance of an inception so that Cobbs could regain his previous life and be able to visit his children. The movie in general received outstanding applause as an innovative and smart story and many critics gave it positive reviews. Rotten tomatoes for instance gave this movie an average score of 8/10 based on over two hundred and fifty reviews and reported that up to 87% of critics gave this movie a positive preview. Specifically praised in this movie is the producer/ director/writer, Mr. Nolan who received many positive notices and compliments even from the best critics. For instance, a notable critic, Peter Travers from the Rolling Stone magazine complimented this movie and Mr. Nolan stating that it was like an ingenious chess game. Justing Chang who said that the writer applied the best skill to vividly depict the procedural detail of sub-consciousness, quite a surrealists thriller which gave all the viewers a big challenge to explore the intricate and deep working of the mind, has praised the writers skill. The genre of this movie is widely interpreted. Some view it as mystery and suspense, while other individuals categorize it as Drama, action, or adventure. Just like any other movie, the issue of genre has been subject to diverse interpretation and thus becoming an element of widespread controversy. The movie Inception correctly falls in the history of science fiction. The first element supporting this fact is that it whole idea of the movie deals with imagined technological or scientific innovations. This also do not happen in the worlds contemporary setting, but a futuristic setting where given the current technological development, such innovations might be possible but the world is not there indeed (George, 2009). Atmans A Semantic/Syntactic Approach to Film Genre, (1984) explores in depth some of the ways used to differential movies into certain genres. He clearly and explicitly explains both the semantic and syntactic approaches to film genres giving us the proper understanding on how to analyze movies and give them appropriate genres. Looking at the work of Altman (1984), this renowned writer observed the same controversy criticism and recognized this approach to genre criticism at semantic approach. In semantic approach, the genre of a certain film is allocated based on some superficial aspects which are thought to fit to that genre. According to this approach, a film is examined based on the plot, characters, and types of some aesthetics among other superficial elements of the movie. Notably about this approach, it falls short of analysis and criticism. This approach fails to look into the details of the movie and less emphasis is laid on the timelines, production and assessment of scenes. The syntactic approach is quite different from semantic approach according to Altman, (1984). This approach takes into account finer details of the movie like the relationships between some semantic elements of that particular genre or between aspects of the society at large and those elements. It appreciates how isolated elements combine in any given movie to come up with the true meaning of such movie (George, 2009). This approach seeks not only to understand why some aspects of any given film are so, but also examines the effects of such aspects on the audience, the information which it deems valuable. This type of film examination brings a deeper look into incidences which might look insignificant in one film but holds a lot of water if compared or connected with similar incidences of other films. Altman goes further to bring up two more different approaches to genre criticism; ideological and ritual. The ritual approach in this sense occurs when moviemakers bent from societal pressures to produce films depicting the contemporary society such as Hollywood. He notes that this type of approach expresses audience desires in broader sense and it has its focus on the consumption side. With the ideological approach, the genre any movie falls into is based on simple, but generalized approaches to identifiable structures (Grant, 2003). Looking at Inception on the light of the Altmans approaches to genre criticism, it can be universally agreed that the same phenomenon is evident. This film clearly supports the arguments made by Altman. The film has been interpreted differently by different authors/different authorities and that is why we have the varying classification in terms of genre. Altman argues that genre is given based on uses and users instead of a fixed point of reference (Grant, 2003). The movie genre is diverse and serves diverse group of individuals. When summing up his arguments, Altman notes that it is not possible to adopt certain terminology that is neutral under any challenge. The terms that are in use are usually based on and derived from different ways for different viewers. It is possible that a certain approach to genre allocation is invalidated by fans regardless of the fact that the studio and nay be the producers validate it. Movies genre is therefore a site of either corporation or struggle among different groups of users or multiple users and such an understanding would help appreciate genre and break away from the tyranny of genre criticism. Altmans arguments are well supported by the film Inception. Different authorities and audiences describe this movie differently. As noted earlier, this movies genre has been describe as fantasy (like in Rotten Tomatoes website), adventure and action, drama, science fiction, suspense and mystery. A casual audience would dismiss this movie as action or drama given the type of violence and gunfights it portrays. If an individual did not pay attention to the overall storyline and did not glimpse the interchanges between real world in the movie, the dreams and dreams within the dreams, it would be obvious that the individual would easily appreciate the action scenes and therefore label the movie action. If some viewers of this movie paid attention to this movies visual effects and the music used in the movie, then it would be easy for such an individual to label it suspense and mystery (Altman, 1984). The visual effects are well set and integrated with the music well to bring out a thrill ing effect of a suspense movie. A careful observer or a person really interested in the movie would note the scientific details about the human mind, dreams and the fiction part on how the ideas are being stolen through the dream. Those who do not understand or appreciate science would label this movie fantasy coming into agreement with Altmans argument that genre just depends on uses, users or groups of users (Altman, 1984). Altman has shade much light on the understanding genre of movies, not only Science Fiction, but all movies at large. Someone may be confused if a movie they clearly know as action movie is referred to as science fiction but if it is an exaggeration of scientific facts, not yet supported by evidence and most likely happens in a futuristic world, such a movie may fall in the science fiction genre. The poem also contains many mythical references and it contains   a great hero.      Ã‚  Ã‚  Ã‚  Ã‚   Beowulf is considered an artifact by many because "it is the oldest of the English long poems and may have been composed more than twelve hundred years ago."(Beowulf 19)   It deals with events of the early 6th century and is believed to have been composed between 700 and 750.   "No one knows who composed Beowulf , or why.   A single manuscript (Cotton Vitellius A XV) managed to survive Henry VIII's dissolution of the monasteries, and the destruction of their great libraries; since his name is written on one of the folios, Lawrence Nowell, the sixteenth-century scholar, may have been responsible for Beowulf's preservation."(Raffel ix)   An interesting fact that is unique about the poem is that "it is the sole survivor of what may have been a thriving epic tradition, and it is great poetry."(Raffel ix)      Ã‚  Ã‚  Ã‚  Ã‚   The poem was composed and performed orally.   "Old English bards, or scops, most likely began by piecing together traditional short songs, called heroic lays; they then gradually added to that base until the poem grew to its present size.   The verse form is the standard Old English isochronic:   each line contains for stresses; there is a strong caesura in the middle of the lines and the resultant half lines are bound together by alliteration.   Although little Old English poetry survives, Beowulf's polished verse and reflective, allusive development suggest that it is part of a rich poetic tradition.

Women's Rights Versus God's Will It seems these days that everyone is concerned with their rights. People feel that there are certain rights that they should have regardless of their race, religion, sex, and so forth. And rightfully so – everyone is entitled to basic human rights. Many belonging to specific groups – such as women, and homosexuals - feel that these basic rights aren't enough and that they need more. They feel that their group should be granted special "rights," which aren't really rights at all. The early advocates of women's rights started out with good intentions – they were fighting for the abolition of slavery, suffrage for women and equal pay for men and women, among other similar rights. The first major turning point in the advocates' battle was on July 19th and 20th of 1848. On those dates, the Seneca Falls Convention was held in New York. The men and women at the convention created a document called the Declaration of Sentiments, which they based on the Declaration of Independence. This document created a foundation, a source of where to go, how to go about getting the government to recognize the rights to which they were entitled. As time went on, woman suffrage advocates like Susan B. Anthony and Elizabeth Cady Stanton formed organizations, such as the NWSA (National Woman Suffrage Association) and the AWSA (American Woman Suffrage Association). Eventually, after the formation of other organizations and after lots of support raising within states, women were granted the right to vote on August 26, 1920. Women's groups didn't stop at getting the right to vote. They worked toward receiving equal pay, equal educational opportunities, and equal job opportunities. People feel that there are certain rights that they should have regardless of their race, religion, sex, and so forth. And rightfully so – everyone is entitled to basic human rights. Many belonging to specific groups – such as women, and homosexuals - feel that these basic rights aren't enough and that they need more. They feel that their group should be granted special "rights," which aren't really rights at all. The early advocates of women's rights started out with good intentions – they were fighting for the abolition of slavery, suffrage for women and equal pay for men and women, among other similar rights. The first major turning point in the advocates' battle was on July 19th and 20th of 1848. On those dates, the Seneca Falls Convention was held in New York. The men and women at the convention created a document called the Declaration of Sentiments, which they based on the Declaration of Independence. This document created a foundation, a source of where to go, how to go about getting the government to recognize the rights to which they were entitled. As time went on, woman suffrage advocates like Susan B. Anthony and Elizabeth Cady Stanton formed organizations, such as the NWSA (National Woman Suffrage Association) and the AWSA (American Woman Suffrage Association). Eventually, after the formation of other organizations and after lots of support raising within states, women were granted the right to vote on August 26, 1920. Women's groups didn't stop at getting the right to vote. They worked toward receiving equal pay, equal educational opportunities, and equal job opportunities.
